Smallpox is one of the deadliest viral diseases in human history, killing an estimated 300 million since 1900 alone. Edward Jenner’s 1796 discovery that skin inoculation with cowpox a scarification method delivery[i][ii] induced immunity against smallpox led to global eradication disease by 1980[iii]. In recent years, vaccinia virus model derived from was used demonstrate (VACVss) superior immunization strategy other methods vaccine delivery[iv]. Relative intradermal or intramuscular injection (i.m.), generates optimal adaptive immunity, indicated VACV-specific CD8+ T memory and IgG titers[v]. The cellular mechanisms behind requirement for have not been elucidated. To test how confers protective we wanted Dendritic Cell (DC) directed modified scarification, focusing on Batf3-dependent[vi] CD103+ dermal DC1s expression DNGR1 death receptor cross-present antigens achieve successful immunity[vii],[viii],[ix],[x],[xi].
